These are the data underlying the figures and analyses presented in Renstrom et al, 2026, Stage-Dependent Effects of “Too Little and Too Much” Medial Prefrontal Activity on Reversal Learning in Rats: Functional Inhibition Impairs Early, Whereas Neural Disinhibition Impairs Late Reversals, J Neurosci.
We examined the impact of bidirectional manipulations of GABAergic inhibition within the medial prefrontal cortex (mPFC) on lever-press reversal performance in male
young adult Lister hooded rats. We used pharmacological functional inhibition of the mPFC by the GABA-A receptor agonist muscimol (MUS) and disinhibition by the antagonist picrotoxin (PTX), and chemogenetic inibition of mPFC GABA neurons by activation of the inhibitory hM4Di DREADD by CNO-2HCl. At every satge we also included a vehicle (saline, SAL) control.
We examined the impact on the spatical discrimination (SD) stage and early reversals (reversals 1-3, R1-3) and on well-established serial reversals (from reversal 5, R5, onwards).
This data set includes the data from the reversal learning experiments, including classical performance measures (responses to criterion, RTC; errors, percentage correct, latencies, omissions), trial-by-trial probabilities of different behavioural strategies (go-previous, go-new, lose-shift, win-shift, sticky), and from the additional studies to characterize expression and functionality of the DREADD in the mPFC, including histological cell counts, baseline-normalized electrophysiological measures, and locomotor activity data.
Data are organised according to how they are presented in the figures of the paper.
